Targeting PIM Kinases to Overcome Therapeutic Resistance in Cancer

چکیده
Abstract Cancer progression and the onset of therapeutic resistance are often results uncontrolled activation survival kinases. The proviral integration for Moloney murine leukemia virus (PIM) kinases oncogenic serine/threonine that regulate tumorigenesis by phosphorylating a wide range substrates control cellular metabolism, proliferation, survival. Because their broad impact on processes facilitate metastasis in many cancer types, it has become clear PIM is significant driver to various types anticancer therapies. As result, efforts target therapy have intensified recent years. Clinical preclinical studies indicate pharmacologic inhibition potential significantly improve efficacy standard targeted This review focuses signaling pathways through which promote therapy, as well highlights biological contexts promising strategies exploit cancer.

Stalling the engine of resistance: targeting cancer metabolism to overcome therapeutic resistance.
Cancer cells are markedly different from normal cells with regards to how their metabolic pathways are used to fuel cellular growth and survival. Two basic metabolites that exemplify these differences through increased uptake and altered metabolic usage are glucose and glutamine. These molecules can be catabolized to manufacture many of the building blocks required for active cell growth and pr...
متن کاملTargeting the Pim kinases in multiple myeloma
Multiple myeloma (MM) is a plasma cell malignancy that remains incurable. Novel treatment strategies to improve survival are urgently required. The Pims are a small family of serine/threonine kinases with increased expression across the hematological malignancies. Pim-2 shows highest expression in MM and constitutes a promising therapeutic target. It is upregulated by the bone marrow microenvir...
متن کاملBcl-2 family proteins in breast development and cancer: could Mcl-1 targeting overcome therapeutic resistance?
Apoptosis, cell death executed by caspases, is essential to normal breast development and homeostasis. Pro-apoptotic and anti-apoptotic signals are tightly regulated in normal breast epithelial cells. Dysregulation of this balance is required for breast tumorigenesis and increases acquired resistance to treatments, including molecularly targeted therapies, radiation and chemotherapies. The pro-...
متن کاملTargeting Notch to overcome radiation resistance
Radiotherapy represents an important therapeutic strategy in the treatment of cancer cells. However, it often fails to eliminate all tumor cells because of the intrinsic or acquired treatment resistance, which is the most common cause of tumor recurrence. Emerging evidences suggest that the Notch signaling pathway is an important pathway mediating radiation resistance in tumor cells. Successful...
متن کاملPIM kinases as therapeutic targets against advanced melanoma
Therapeutic strategies for the treatment of metastatic melanoma show encouraging results in the clinic; however, not all patients respond equally and tumor resistance still poses a challenge. To identify novel therapeutic targets for melanoma, we screened a panel of structurally diverse organometallic inhibitors against human-derived normal and melanoma cells. We observed that a compound that t...
